What’s the mechanism of aging? The Sirtuin family members are renowned for their roles in regulating lifespan. Out of the seven mammalian Sirtuins, it is Sirt6 that plays the most critical role. Sirt6 whole-body overexpression has been rigorously shown to extend mice lifespan 15 to 27 percent depending on the gender and strain, and Sirt6 whole-body knockout mice can’t survive over one month. As long as its molecular activities, cellular functions and their physiological relevance are fully comprehended, we will get closer to unveil the mystery. In this lab, my project is to characterize a Sirt6 separation of function mutant mice.
